چکیده

Plasma membranes from bovine corpora lutea have been purified by sucrose density gradient centrifugation. The purified membranes, in addition to binding 1251-hCG, also possess hCGstimulated adenylate cyclase and Na-K-ATPase. The relative purification of 1251-hCG binding, adenylate cyclase and Na-KATPase on the basis of the specific activities in the whole homogenate were 7.8, 6.4 and 2.6, respectively. The presence of both the hormone sensitive adenylate cyclase and 1251-hCG binding activities suggest that these plasma membranes might possess the 'receptor' for gonadotropin.

Isolation of the luteinizing hormone-chorionic gonadotropin receptor in high yield from bovine corpora lutea. Molecular assembly and oligomeric nature.

The luteinizing hormone-human chorionic gonadotropin (LH-hCG) receptor was isolated from 8-liter batches of a supernatant (7,000 X g) of bovine corpora lutea homogenate from 1,200 ovaries obtained biweekly from the Wampole Laboratories, Princeton, NJ. The supernatant was concentrated by ultrafiltration to remove an inhibitor of hormone-receptor binding and inert proteins of M, < 50,000. The con...

Comparison of heart sarcolemmal enzyme activities in normal and cardiomyopathic (UM-X7.1) hamsters.

1. The activities of some membrane-bound enzymes such as adenylate cyclase, Na+ + K+-stimulated adenosine triphosphatase (Na+ + K+-ATPase), Ca2+-stimulated ATPase and Mg2+-stimulated ATPase were examined in heart sarcolemmal fractions from control and cardiomyopathic hamsters at different stages of heart failure. 2. The basal adenylate cyclase activity in sarcolemma from cardiomyopathic animals...

In vitro examination of LH-HCG receptors in human corpora lutea of the menstrual cycle and pregnancy.

Binding of 125I-human chorionic gonadotropin (HCG) to homogenates of the human corpora lutea of the menstrual cycle and pregnancy was examined in vitro. While corpora lutea of the menstrual cycle bound 125I-HCG specifically, the corpora lutea of pregnancy from 8 weeks' gestation bound little or none of the added 125I-HCG. In the corpus luteum of the menstrual cycle the dissociation constant for...

Distribution of enzyme activities in subcellular fractions of bovine retina.

Centrifugation of homogenates of bovine retinas to isopycnic equilibrium in sucrose density gradients yielded three partially overlapping bands of particles which were, in the order of increasing density: (a) photoreceptor cell (rod) outer segments; (b) plasma membranes, lysosomes, and large fragments of endoplasmic reticulum; and (c) mitochondria. The only enzyme activity investigated which ha...

Properties of gonadotropin receptors in the cell membranes of bovine corpus luteum.

The isolated cell membranes from bovine corpora lutea bound lz51-human chorionic gonadotropin (HCG) with high affinity and specificity. The specific binding was saturable with respect to the amounts of membrane protein and 12jIHCG added. Approximately 8.4 x 10-l” M 124-HCG saturated all the receptor sites in 1.0 mg of membrane protein. The specific binding of 12sI-HCG was strong and not readily...
